Mutations in the APC and MUTYH genes

FAP MASTR™ is a molecular assay for the identification or confirmation of the presence or absence of mutations in the complete coding regions of the APC and MUTYH genes. This test is ready to use and offers robust performance with minimum hands-on time. It consists of all the reagents necessary to enable multiplex amplification of 52 amplicons (245-455 bp) for complete exon coverage of the APC and MUTYH genes. Only three Multiplex PCR reactions are necessary to perform the test. In addition, the FAP MASTR™ is compatible with all current Massively Parallel Sequencing (MPS) platforms.

Detection of variants during amplicon resequencing is very important. It enables sequencing errors to be distinguished from real variants, and consequently greatly improves data accuracy. Single natural nucleotide addition sequencing technologies such as 454 pyrosequencing and Ion Semiconductor sequencing are used to detect variants, but one of their limitations is to differentiate between stretches of identical bases (i.e. homopolymers) of 6 bp or higher. Because HP mutations will result in frameshift changes within characterized disease genes, Multiplicom’s HP assays are the ideal tests to detect homopolymer stretches that are difficult to detect with current single natural nucleotide addition MPS platforms.
