MID for Ion PGM™ System - Multiplicom

For the second PCR to incorporate barcoding and adaptors

These kits are to be used in combination with the MASTR™ assays for the second PCR to incorporate barcoding and adaptors required for the Ion PGM™ System (Ion Torrent/Thermo Fisher Scientific).

In this second PCR (i.e. Universal PCR) the MASTR-derived amplicons are tagged resulting in the incorporation of an MID and the A and P1 adaptors required for sequencing on the MPS System. For each PCR Mix two separate reactions are performed, to allow bidirectional sequencing of the amplicons. One kit contains 8 specific MID Primers mixes.

Reagents for "Universal PCR": 24 dual PCR reactions per MID (1-8), 192 reactions in total.

Reagents for GeneScan labeling, 192 reactions in total.

The MID for Ion PGM™ System kits are to be used for research purposes only.  Currently, the MID for Ion PGM™ System kits have been verified for amplification and sequencing coverage of the genomic targets in the CFTR MASTR Dx, FMF MASTR Dx, EGFR 18-24 MASTR and SOMATIC 1 MASTR.  The performance for variant calling using any of these MASTR assays for sequencing with the Ion PGM™ System must be validated. When using the MID for Ion PGM™ System kits in combination with a CE-marked MASTR™ Dx kit* for diagnostic purposes, the combined workflow should first be validated. 

* For BRCA MASTR Dx, our Short Read Amplification kit followed by Life Technologies' library preparation steps is the only method for accurate sequencing according to their “Decision Tree for DNA Sequencing on the Ion PGM™ System” and must be validated for diagnostic purposes.
