For the detection of homopolymer variation using capillary electrophoresis

Multiplicom has developed homopolymer assays for each of their MASTR™ assays to allow rapid detection of small insertion/deletion mutations in the coding homopolymer stretches of the targeted genes since these mutations are difficult to detect via some Massive Parallel Sequencing platforms. All the reagents necessary for rapid detection of changes in the length of coding homopolymer stretches are included to amplify the targeted fragments using multiplex PCR.

Detection of variants during amplicon resequencing is very important. It enables sequencing errors to be distinguished from real variants, and consequently greatly improves data accuracy. Single natural nucleotide addition sequencing technologies such as 454 pyrosequencing and Ion Semiconductor sequencing are used to detect variants, but one of their limitations is to differentiate between stretches of identical bases (i.e. homopolymers) of 6 bp or higher. Because HP mutations will result in frameshift changes within characterized disease genes, Multiplicom’s HP assays are the ideal tests to detect homopolymer stretches that are difficult to detect with current single natural nucleotide addition MPS platforms.
