Mutations in exons 9, 11, 13, 14, 15, 16, 17 of the KIT gene and exons 8, 10, 12, 14, 18 of the PDGFRA gene

The GIST MASTR assay amplifies exons 9, 11, 13, 14, 15, 16, 17 of the KIT gene and exons 8, 10, 12, 14, 18 of the PDGFRA gene. The assay contains 17 amplicons (120-220 bp), which are amplified in 2 multiplex PCR reactions.

Gastrointestinal stromal tumors (GISTs) are the most common forms of connective tissue tumors or sarcomas and occur in approximately 20 per one million people adults between ages 40 and 70.

Roughly 80% of GISTs are associated with a mutation in the KIT gene, and about 10 % are associated with a mutation in the PDGFRA gene. Mutations in KIT and PDGFRA are associated with both familial and sporadic GISTs. The KIT and PDGFRA genes both code for tyrosine kinase receptors that if mutated no longer require ligand binding to be activated. As a result, the proteins and the signaling pathways are constantly turned on (constitutively activated), leading to uncontrolled cell growth resulting in GIST formation.

The most common treatment for GIST is surgery in combination with targeted treatment with tyrosine kinase inhibitors.
