Join Dr. Alexis Gonzalez as he presents results that are part of an ongoing study by researchers at the University of Melbourne on the cellular distribution and biological role of a barely known innate immunity surface receptor. Guaranteeing success in this challenging cytometry project, including the unbiased detection of very dim tdTomato tag on cells of varying autofluorescence, called for a multi factor approach: Identifying the best instrument for the job in terms of sensitivity; a careful design of the multi-color panel to minimize unmixing/compensation-derived spreading; and the use of several strategies for the isolation and extraction of spectrally unique autofluorescence sources. The results reveal the power of Aurora autofluorescence extraction in the detection of dim fluorescence on highly autofluorescent cells, and the clear advantage this feature provides to Aurora over highly sensitive instruments that lack the autofluorescence extraction feature or the capacity to capture intrinsic autofluorescence in fine detail.
